        <title id="abstract-title-4825ce26a6584e36bbd38a432c6d9d36">Abstract</title>
        <p id="paragraph-5c6f1a30a871417ba2423d0f96f5d7d0">Psoriasis is recognized as the most common autoimmune disease caused by the inappropriate activation of the cellular immune system, characterized, by severe skin inflammation, epidermal hyperproliferation, the lack of possible cure and associated severe side effects in allopathic medicines has led to extensive research in natural products with antipsoriatic activity. <italic id="e-0c37d1d60768">Aloe sinkatana</italic> plant is a very potent antipsoriatic agent and is used traditionally to treat psoriasis. To formulate and evaluate a new emulgel from <italic id="e-f7006c3d1dac">Aloe sinkatana</italic> plant extract used as an anti-psoriatic agent. A 2³ factorial design was selected to formulate <italic id="e-1f4a74b6ec38">Aloe sinkatana</italic> Eumlgel. The three factors were: the amount of gelling agent X1, amount of emulsifying agent X2, and amount of liquid paraffin X3, these were selected as independent formulation variables, while the viscosity and spreadability were taken as dependent variables or response variables. The model of the formulation was considered statistically excellent with R² values approaching unity since, the R² values for both the response's viscosity and spreadability were found to be 0.9915, and 0.9761 respectively. The extract of <italic id="e-e7d455f22020">Aloe sinkatana</italic> was formulated as an active ingredient in the emulgel dosage form, and formula F3 was selected as an optimal emulgel formula.</p>

      <title id="title-1d47bbcbd4bc429e93840d7cc363741b">INTRODUCTION</title>
      <p id="paragraph-ba43a71e95564296a87e58b0c69410ef">Psoriasis is recognized as the most common autoimmune disease caused by the inappropriate activation of the cellular immune system. The National Psoriasis Foundation reports that this disease affects approximately 125 million people worldwide (2% to 3% of the total population have psoriasis) <xref id="xref-acaab055b965485b91473cb7a69c75a5" rid="R264918833010610" ref-type="bibr">1</xref>. Nearly 60% of people with psoriasis reported their disease to be a large problem in their everyday life. Total healthcare costs of psoriasis for patients are calculated at $11.25 billion annually, approximately 60% of psoriasis patients miss an average of 26 days of work a year due to their illness <xref id="xref-7170dac9c61d434bbb6ac35ad1831f01" rid="R264918833010596" ref-type="bibr">2</xref>. The current available synthetic drugs used to treat psoriasis have not fully met the needs of the sufferers, largely due to several problems like resistance to treatment after long-term drug exposure and relapses upon cessation of medication after partial or acceptable clearance is obtained. Also, some treatments may increase the risk of cancer (phototherapy) and can induce disorders in the liver <xref id="xref-dcfff6a1367046a9beb9bc9e61b9c66a" rid="R264918833010612" ref-type="bibr">3</xref>. Furthermore, the treatment efficacy may diminish with time, and it must be replaced by another therapy. Therefore, at present, there is still no curative treatment for psoriasis. </p>
      <p id="paragraph-19da7c7079a34f50a034a29caecf2836"><italic id="e-91c394d68fe4">Aloe sinkatana</italic> plant is a very potent antipsoriatic medicinal plant that grows naturally in the Eastern Sudan in the Red Sea Mountains mainly in the Sinkat area, where it is popularly used extensively by residents of the region to treat psoriasis <xref rid="R264918833010608" ref-type="bibr">4</xref>, <xref rid="R264918833010600" ref-type="bibr">5</xref>. Bearing in mind all this, the main task of this study is to formulate and evaluate a new safe and effective emulgel from <italic id="e-dbeae3a2b4b7">Aloe sinkatana</italic> plant extract used as an anti-psoriatic agent.</p>
      <p id="paragraph-4e708c97ef22477cab57c8d779c5bbac">Emulgel is topical drug delivery, and to date, it has less marketed product, so it is interesting and challenging to focus on emulgel. It is a combination of gels and emulsions. </p>
      <p id="paragraph-d4b3bb54be23436f93d98549c643ca20">Emulsions are controlled release systems containing two immiscible phases in which the internal phase is dispersed into external, with the use of an emulsifying agent to stabilize the system. Emulsions are of oil-in-water or water-in-oil type, where the active agent particle entrapped in the internal phase passes through the external phase,<xref rid="R264918833010594" ref-type="bibr">6</xref>, <xref rid="R264918833010605" ref-type="bibr">7</xref> USP defines gel as a semisolid system consisting of dispersions made up of either small inorganic particles or large organic molecules enclosing and interpenetrated by liquid. The gel contains a larger amount of aqueous liquid in a cross-linked network of colloidal solid particles where it captures small drug particles and maintains the controlled release of the drug. The emulsion and gel both are responsible for the controlled drug release from the systems <xref id="xref-c6047d0ad5e94a6287f3f2fa2d24d4aa" rid="R264918833010614" ref-type="bibr">8</xref>, gels have still limitations in the delivery of hydrophobic drugs so to overcome this limitation the concept of emulgel was introduced where the hydrophobic drugs are incorporated in emulsion and then to gel <xref rid="R264918833010597" ref-type="bibr">9</xref>, <xref rid="R264918833010606" ref-type="bibr">10</xref>. Emulgel combines the advantage of emulsion and gels, gaining the dual controlled release effect, where the emulsion is gelled by incorporation in the gel base <xref id="xref-ca0cbadc70f146a4a3e7a8acb2b3f23d" rid="R264918833010601" ref-type="bibr">11</xref>, emulgel shows good stability and better release of drugs, also it can be used to prolong the effect of drugs having shorter t1/2. Furthermore, materials used in the Preparation of emulgels are available, and cheaper, resulting in, a reduction in emulgels production costs <xref rid="R264918833010616" ref-type="bibr">12</xref>, <xref rid="R264918833010595" ref-type="bibr">13</xref>, <xref rid="R264918833010603" ref-type="bibr">14</xref>.</p>
      <p id="paragraph-db9760373ddf45d5b10e905f600656a1">The most important constituent of emulgels is an aqueous phase (water, alcohol, etc), oil phase (eg mineral oils liquid paraffin, propylene glycol, isopropyl myristate, isopropyl palmitate, castor oil, olive oil, balsam oil, wool wax, soyabeen oil, cotton seed oil, oleic acid, maize oil, arachis oil, etc.), emulsifying agent (e.g. Polyethylene glycol, Span 80, Tween 80, Stearic acid, Sodium stearate, etc.), gelling agent (e.g. Carbopol 934, 940, xanthan gum, etc.) and permeation enhancer (e.g. Oleic acid, Menthol, Clove oil, Lecithine, Isopropyl myristate, Urea, Linoleic acid, Cinnamon, etc.) <xref rid="R264918833010613" ref-type="bibr">15</xref>, <xref rid="R264918833010604" ref-type="bibr">16</xref>, <xref rid="R264918833010607" ref-type="bibr">17</xref>. </p>

    <sec>
      <title id="title-0ec0866b77ec4b65851fff7d01cb3064">MATERIAL AND METHODS</title>

        </title>
        <sec>

            <bold id="strong-686f7f0b93f544abae9758700b418662">The standard compound</bold>
          </title>
          <p id="paragraph-a190e30e85074c619eb1fa70f82bac15">Aloe-Emodin standard compound was received as a gift from Prof. Masaki, (Kobe University, Japan). </p>
        </sec>
        <sec>
          <title id="t-4d3a3c0dbe3e">
            <bold id="strong-217cd7969291408fb0f3045022fdbbdd">Plant Collection and Authentication</bold>
          </title>
          <p id="paragraph-5d0d3450fb154fa6ad83165130686ce8">The <italic id="e-d65b93e88970">Aloe sinkatana</italic> plants were collected, from the Sinkat area, Red Sea state, Sudan, they were authenticated by the Department of Chemistry, Medicinal &amp; Aromatic Plants Research Institute and Traditional Medicine, National Research Center, Khartoum, Sudan.</p>
        </sec>
      </sec>
      <sec>
        <title id="t-75c81b281d86">Methods</title>
        <sec>
          <title id="t-3fee258b22c0">
            <bold id="strong-4dbba6fde62e405bb9c1f9bef1cdedfa">Extraction of <italic id="e-ef256a1c4f8b">Aloe </italic></bold>
            <italic id="e-ef256a1c4f8b-e18d8216-e17c-447e-8206-d04af403aa75">
              <bold id="strong-b175cc67495340faac4de2e9fad0657b">sinkatana</bold>
            </italic>
          </title>
          <p id="paragraph-4062ec826a4d4ea9882b633719136a0c">Mature, healthy, and fresh leaves of <italic id="e-aa389189c981">Aloe sinkatana</italic> having a length of approximately 75 to 90 cm were washed with fresh water. </p>
          <p id="paragraph-0f1da4406d2247758007a6be8b81517e">The inner gel is scrapped and cut into pieces. A traditional hand filleting method of processing Aloe leaves was used. In this method, the lower leaf base, the tapering point at the leaf top, and the short spines located along the leaf margins were removed by sharp blades. The blade was then introduced into the mucilage layer below the green rind avoiding the vascular bundles, and the top rind was removed. The epidermis of the leaves was peeled off, and the colorless, solid mucilaginous gel was cut into pieces. Then 250 gm of gel was loaded into a 1000ml flask and 500 ml solvent (Ethanol) was added. Ultrasound-assisted extraction was performed at 60° C for 60 min. After that the solution was filtered and the solvent was removed under reduced pressure in a Rotary evaporator until it became completely dry <xref id="xref-725fffdef97b496080e5f9762247473c" rid="R264918833010607" ref-type="bibr">17</xref>. </p>
        </sec>
        <sec>
          <title id="t-a2d2cbcbd35c">
            <bold id="strong-db74baf403d44c57b28e39ea113d81d7">Formulation design using 2³ full factorial design</bold>
          </title>
          <p id="paragraph-a7f6f813bbd64995a91d9bb839e2eedd">To obtain the "best" or an "optimized product" eight different formulations were generated using two levels, three-factor, full factorial design.</p>
          <p id="paragraph-a647beaf0b2a4a73989dc0ee3a52008e">A 2³ factorial design for three factors at two levels each was selected to optimize the varied response variables. The three factors, amount of gelling agent X1, amount of emulsifying agent X2, and amount of liquid paraffin X3 were selected as independent formulation variables, and the factor levels were suitably coded (<xref id="x-c2df42c708ae" rid="table-wrap-c446022320b34a3993767d7f7aaa0855" ref-type="table">Table 1</xref>). Viscosity and Spreadability were taken as dependent variables or response variables. Experimental trials were performed at all 8 possible combinations (<xref id="x-11e4ae58ab98" rid="table-wrap-34aeb180f19547828ebcd1885bcf130a" ref-type="table">Table 2</xref>). All other formulation variables and processing variables were kept invariant throughout the study <xref id="xref-df926c4b9e8948d388da8507718d37f0" rid="R264918833010599" ref-type="bibr">18</xref>. </p>
          <p id="paragraph-ef63221b0ed44b88ac0f541e8f50b5c2">Design-Expert_DX 8 Software was used for the generation and evaluation of the statistical experimental design. </p>

            <bold id="strong-03b6a5e20ee44df0bb524588b86b4c76">Emulgel</bold>
          </title>
          <p id="paragraph-0fcaafeaa1f14a81923e2a8b5e3a1a9b">The composition of emulgel formulations is shown in <xref id="x-5adef213d14f" rid="table-wrap-3360dc7902c94f87a3633ab6b46cff22" ref-type="table">Table 3</xref>.</p>
          <p id="paragraph-6bbd3b1c7a6d4b6c8c626abec21c63bb">First, the gel was prepared by dispersing Carbopol 934 in heated purified water (80 °C), and the dispersion was cooled and left overnight. The oil phase of the emulsion was prepared by dissolving Span20 in liquid paraffin while the aqueous phase was prepared by dissolving Tween20 in purified water. </p>
          <p id="p-98b552317d71">Propylene glycol was dissolved in ethanol whereas plant extract was dissolved in DMSO, and both solutions were mixed with the aqueous phase. Both the oily and aqueous phases were separately heated to 70 to 80 °C then the oily phase was added to the aqueous phase with continuous stirring until cooled at room temperature. The obtained emulsion was mixed with the gel in a 1:1 ratio with gentle stirring to obtain the emulgel. Finally, the pH of the emulgel was adjusted by using triethanolamine <xref rid="R264918833010611" ref-type="bibr">19</xref>, <xref rid="R264918833010609" ref-type="bibr">20</xref>.</p>
        </sec>
        <sec>
          <title id="t-19e75c297494">
            <bold id="strong-332059b133d64bb7857b9c0f782bafe5">Macroscopic evaluations</bold>
          </title>
          <p id="paragraph-122de10b389d4bb4ac628f1c18d10163">The 8 batches of the emulgel formulae were viewed under a microscope to study their color, texture, homogeneity, consistency, and phase separation <xref rid="R264918833010815" ref-type="bibr">21</xref>, <xref rid="R264918833010813" ref-type="bibr">22</xref>.</p>
        </sec>
        <sec>
          <title id="t-72af0fcc25d9">
            <bold id="strong-74082b083c4145d6aec7c4b793c23082">Centrifugation study</bold>
          </title>
          <p id="paragraph-32d9e53a4fd54111a6bef6f42ab53d9a"> Centrifugation study is a useful means to check the stability of the prepared emulgel formulae. These studies were performed one week after the preparation of emulgels. Centrifugation was carried out using a Mini centrifuge at 3000 rpm for 30 minutes <xref id="x-969d9c9804b2" rid="R264918833010816" ref-type="bibr">23</xref>.</p>
        </sec>
        <sec>
          <title id="t-ff8a25898fa3">
            <bold id="strong-0abe9c5b400e4018ab2c4264bd9f3270">Measurement of pH</bold>
          </title>
          <p id="paragraph-9cc862b54f9f47cd812650aa63245a64">The pH value of 1% aqueous solutions of the prepared <italic id="e-3ff5d745f623">A. sinkatana</italic> emulgel was measured using a pH meter (Max Instruments Chandigarh, India), which was calibrated before each reading, with buffered solution at pH4.0 and 7.0. The measurement of the pH of each formulation was done in triplicate and average values were calculated. <xref rid="R264918833010819" ref-type="bibr">24</xref>, <xref rid="R264918833010818" ref-type="bibr">25</xref>, <xref rid="R264918833010817" ref-type="bibr">26</xref> </p>
        </sec>
        <sec>
          <title id="t-87a9005b24f5">
            <bold id="strong-35ab31cf794f4dc48f8fc2fb2585632c">Viscosity Test</bold>
          </title>
          <p id="paragraph-d50b65d98e714b7cbe0f969d893f85d6">Measurements of viscosity were done by Viscometer VR 3000 (Viscotech, Spain) by choosing the appropriate spindle number (L4) and rpm (100 rpm) at room temperature. An appropriate amount of each emulgel formulation was kept in a suitable beaker, the spindle groove was dipped, and the rpm was set. Viscosity measurements were started, and the readings were measured after 1 minute, and the viscosity of each formulation was calculated <xref rid="R264918833010820" ref-type="bibr">27</xref>, <xref rid="R264918833010822" ref-type="bibr">28</xref>.</p>
        </sec>
        <sec>
          <title id="t-f0320e585842">
            <bold id="strong-a5b6bfc3bdca45098e8d3066fd30690b">Spreadability</bold>
            <bold id="strong-bdffe11a9c9e405093a3cc3e4eedade3"> test</bold>
          </title>
          <p id="paragraph-5555071d4985410a92b96bfabfea4aec">The Spreadability was determined by the parallel plate method, which is widely used, by measuring the spreading diameter of 1gm of emulgel between two horizontal plates (30cm x 30cm) after one minute <xref id="x-0f4f9a16abbb" rid="R264918833010821" ref-type="bibr">29</xref>.</p>
        </sec>
        <sec>
          <title id="t-4e95d181b23a">
            <bold id="strong-19ab554428f348e187d970d2729f4837">Determination of the emulsion type formed (Dilution test)</bold>
          </title>
          <p id="paragraph-477adba982914a08873d2756b34bb006">The emulgel formulae were diluted either with oil or water. If the emulsion of the emulgel is o/w type and it is diluted with water, it will remain stable as water is the dispersion medium but if it is diluted with oil the emulsion will break as oil and water are not miscible with each other <xref id="x-334aca316d85" rid="R264918833010824" ref-type="bibr">30</xref>.</p>
        </sec>
      </sec>
      <sec>
        <title id="t-41939a0be759">
          <bold id="strong-c467092b6f2a43dfa9e942edaaaf57eb">Quantitative analysis</bold>
        </title>
        <sec>
          <title id="t-523f99155dfe">
            <bold id="strong-ba20cf70e9ac4e258a02878dbfef513b">Calibration curve of standard Aloe-emodin compound </bold>
          </title>
          <p id="paragraph-f6553414204843d980edbff16a3d3707">Accurately weighed Aloe-emodin standard material (0.01g) was, transferred to a 100 ml volumetric flask and dissolved by ultrasonic shaker with methanol to get a standard solution of 100µg/ml (stock solution). Various concentrations (2 µg /ml, 4 µg / ml, 6 µg /ml, 8 µg /ml, 10 µg /ml, and 12 µg /ml) of stock solution were made and the absorbance of various dilutions was taken at wavelength 430 nm using a UV spectrophotometer. The calibration curve was determined by plotting the absorbance, against the concentration.</p>
        </sec>
        <sec>
          <title id="t-06abf8f68517">

          <p id="paragraph-6c9d205ad97f40a8a5a251ac53890d81">0.5gm of dried extracts were dissolved by ultrasonic shaker in 100ml of methanol (0.5%). The absorbance of the resultant solution was read at wavelength 430 nm using a UV spectrophotometer.</p>
        </sec>
        <sec>
          <title id="t-e01c6b3f02b9">
            <bold id="strong-7f0206600755432a89f79e2fbb8f3db8">Determination of Drug Content</bold>
          </title>
          <p id="paragraph-61a4f920e4fb4689b1dd7c9a5ad3be26">For determination of the aloe emodin compound content in the different formulas of <italic id="e-f27298694d77">Aloe sinkatana</italic> extract was weighted 5gm of emulgel formula and dissolved by ultrasonic shaker in 50ml of methanol (10%). The absorbance of the resultant solution was read at wavelength 430 nm using a UV spectrophotometer. The percentage drug content was calculated.</p>
        </sec>
        <sec>
          <title id="t-363024cf9037">
            <bold id="strong-5de51d67ebb24b349212fc27bbb0211b">Statistical analysis</bold>
          </title>
          <p id="paragraph-830d60e04fd94b59a265a7fb6d214844">The data obtained was statistically analyzed by Design-Expert_ DX 8 Software statistical program.</p>
        </sec>

      <p id="paragraph-31c8028e25404c52bebcb40372b59002">The emulgel formulas are glossy, reddish brown in color; all formulations are homogeneous, stable, and smooth in texture, as shown in <xref id="x-a1bcae1190e3" rid="table-wrap-94d31dc1fd384b7c9557cd93d4a6486c" ref-type="table">Table 4</xref>.</p>
      <p id="paragraph-c120f3a37dfd47b28d833084374bcc7d">The centrifugation test, measured pH, viscosity and spreadability values of the prepared emulgel formulations are given in <xref id="x-abd89acc7e7b" rid="table-wrap-5d681ad5649f41f3ab460faf78bb6a14" ref-type="table">Table 5</xref>. Centrifugation is an important tool for evaluating and predicting the shelf life of semisolid formulations like emulsion. The centrifugation test (stress testing) is usually used to evaluate the physical stability of semisolid formulation stored at different temperatures in terms of phase separation <xref id="xref-887591dabbc04048b5d4eff3758b8574" rid="R264918833010602" ref-type="bibr">31</xref>, the results show that all the formulas maintained their consistency and homogeneity after 30 minutes of centrifugation and no phase separation was detected, except the formula F5 showing phase separation after 25 minutes of centrifugation and this may be due to the low concentration of emulsifying agent and gelling agent </p>
      <p id="paragraph-33daf57490324f22a134c91d210283bc">The pH values of the formulations range from 6.36 to 6.48, which is considered acceptable to avoid the risk of irritation upon application to the skin (within the pH range of the human skin). This was achieved by the incorporation of triethanolamine. The physiologic role of an acidic skin surface historically was thought to be a defense mechanism against invading organisms. More recently, it has been demonstrated that several key enzymes involved in the synthesis and maintenance of a competent skin barrier are largely impacted by pH. Hence, a broader view of the importance of pH in the function and integrity of the skin is emerging <xref id="xref-e3dc2bd2e529412d8c555dbf7a81b5b5" rid="R264918833010615" ref-type="bibr">32</xref>.</p>
      <p id="paragraph-d37eb75dce18415fa644b7b3436ac79c">Results of the viscosity test showed that the viscosity increased as the amount of gelling agent and a surfactant increased and vice versa, the viscosity ranged between 26734 to 28510 Cps. While the results of spreadability ranged between 23.2 to 25.1cm. </p>
      <p id="paragraph-54f29ed24f004db4a4995a568e9f525c">To determine the emulsion type, all prepared formulas were diluted with oil and there was no phase separation upon dilution (stable), however, when water was used for dilution the emulgel of all formulas was separated into two phases, and this result indicated that the emulsions were W/O type.</p>
      <p id="paragraph-a37530b73fba47d2924087c7e36b1d13">The absorbance for the concentration of Aloe Emodin standard compound is shown in <xref id="x-b5eecf8807ad" rid="table-wrap-ef355637de54424aaa0660ed3182a56d" ref-type="table">Table 6</xref>, and the calibration curve is illustrated in <xref id="x-1e9a50bab945" rid="figure-ef12a7e4dbf442eabe15f6a475a13deb" ref-type="fig">Figure 1</xref>. The standard graph of Aloe Emodin compound shows good linearity with an R2 value of 0.986, which indicates that it obeys Beer's-Lambert's Law in the concentration range of 0-100 µg/ml. </p>
      <p id="paragraph-3cf8e2b3e1a24603ac7043f7738d4270">The aloe-emodin concentration in <italic id="e-b3d9c8fed270">Aloe sinkatana</italic> extract and then the drug content was estimated from <xref id="x-05b02dd01d1a" rid="disp-formula-group-f3a3a6be520d496d899a4dddeaa09ac6" ref-type="disp-formula">Equation 1</xref> of the calibration curve. The final concentration of Aloe-emodin in the plant extract was found to be equal to 4 µg /5mg = 0.08%</p>
      <p id="paragraph-22375063e2ae4e00a0582a979873ca4e">The result of drug content of the 8 formulae ranged between 92.34% &amp; 99.48, while the (F3) formula shows the highest percent of drug content (<xref id="x-22ebe257884a" rid="table-wrap-553e1a38882c4bf3b31482a6c5439469" ref-type="table">Table 7</xref>).</p>
      <p id="paragraph-7cad0041666f4928b7d03993e92c83a6">Analysis of variance (ANOVA) according to the data of the responses (Y1&amp;Y2) for both viscosity and spreadability is shown in <xref rid="table-wrap-9971669ad2024eca875b2cdd62fc95d3" ref-type="table">Table 8</xref>, <xref rid="table-wrap-c2c813ade295499fb2af849f3724509f" ref-type="table">Table 9</xref>. All P-values listed on <xref rid="table-wrap-9971669ad2024eca875b2cdd62fc95d3" ref-type="table">Table 8</xref>, <xref rid="table-wrap-c2c813ade295499fb2af849f3724509f" ref-type="table">Table 9</xref>, indicated a significant effect of the independent factors (X1, X2 &amp; X3) on the responses (Viscosity and Spreadability). The linear model was selected for both responses (viscosity and spreadability) with Model F-value 151.88, P- value is 0.0001 and F-value 54.54, P- value is 0.0011, respectively. </p>
      <p id="paragraph-039e78494f22460fb79b6a78d8ec8d02">The goodness of fit of the model was considered statistically excellent with R2 values approaching unity. The values for the responses Y1, and Y2 were found 0.9915 to 0.9761. Therefore, the model was found statistically excellent for both responses Y1 and Y2.</p>
      <p id="paragraph-60b9582a27124237be7e413c62e20e49">To validate the predictive ability of the hypothesized model for each response, the agreement between predicted and observed responses was verified; the experimental and predicted values were within the confidence interval for each response, the "Predicted R squared" of 0.9652 is in reasonable agreement with the "Adjusted R Squared" of 0.9848 in case of viscosity while the value "Predicted R squared" of 0.9652 is in reasonable agreement with the "Adjusted R Squared" of 0.9582in case of spreadability. These results indicate that the viscosity and spreadability both are strongly affected by the variables selected for the study. Consequently, we can conclude that the statistical model is mathematically valid. The resulting equations for dependent variables—Y1 (Viscosity), and Y2 (spreadability) in terms of coded factors are presented below.</p>

      <p id="paragraph-244829afe25145a089fb9ad2d8066d01">The regression <xref rid="disp-formula-group-48368d0024f4417a98b07be6bf4a8b50" ref-type="disp-formula"/>, <xref rid="disp-formula-group-5d7f494b5c204273aaf075b0dd458d14" ref-type="disp-formula"/> presented in <xref rid="figure-1fd1184c119f44188e7bf0057fb2dc2f" ref-type="fig">Figure 2</xref>, <xref rid="figure-645abfeeaf204fad91a968bb6723693f" ref-type="fig">Figure 3</xref> show the influence of independent variables X1, X2, and X3 on the viscosity (Y1) and spreadability (Y2). It was clear that the three independent variables had a significant influence on Y1 and Y2 but the most important (highest) effect occurred by factor X1 which is the amount of gelling agent (P- value &lt; 0.0001 &amp; 0.0003 respectively).</p>
      <p id="paragraph-6436ce22180f4b6ba54cd8a9ccd04ee3">Both variables (amount of gelling agent -X1 and amount of surfactant-X2 have a positive effect (synergistic effect) on viscosity and a negative effect (antagonistic effect) on the spreadability. The variable (amount of liquid paraffin -X3) has a direct relationship with spreadability and an inverse relationship with viscosity, that mean when viscosity is very high, the formulation becomes rigid and the spreadability decreases. So, it is necessary to choose the optimum amount of variables to get good viscosity. Formulation variables, e.g. gelling agent, surfactant, emulsifying agent, and oil phase influence the rheological properties of the formulation.</p>
      <p id="paragraph-f5b3e7d5cb56411da054ccb9f30d7733">The model graph for the viscosity is given in <xref id="x-14c376086a4f" rid="figure-1fd1184c119f44188e7bf0057fb2dc2f" ref-type="fig">Figure 2</xref>. The figure showed the viscosity increased as the amount of a gelling agent and a surfactant increased and vice versa. </p>
      <p id="paragraph-9f4ed45d7c5a437aafa185cb86f3aade">The graphical result in <xref id="x-4752d74add72" rid="figure-645abfeeaf204fad91a968bb6723693f" ref-type="fig">Figure 3</xref> showed that the spreadability is indirectly proportional to the amount of a gelling agent and a surfactant, and directly proportional to the amount of liquid paraffin (linear model). </p>

    <sec>
      <title id="title-f0b6a6d408384b25921c6412442d0255">Conclusion</title>
      <p id="paragraph-573bdd68c6334961ae20ba90bab8b263"> Aloe-emodin has been determined as one of the active constituents of <italic id="e-c0f8b7a60bac">Aloe sinkatana</italic> as a marker compound. The antipsoriatic emulgel has been formulated from <italic id="e-605451801b61">Aloe sinkatana</italic> extract using 2³ factorial design which fulfills all standard requirements. It was finally concluded that the developed formulation F3 was found to be a more promising topical herbal emulgel as it shows good physicochemical characteristics highest percent of drug content. Future Research works can be done to get better results.</p>
